新近,用同种异体混合白细胞反应进行的研究表明,Leu-3~+T细胞中的Leu-8~+和Leu-8~-亚群在体外细胞介导免疫反应中起重要作用.为了确定它们是否参与体内细胞介导免疫反应,以及证实多种细胞HLA-DR、HLA-DQ的表达、T细胞Tac和Ki-67抗原的表达、郎格罕细胞Leu-3的表达情况,作者用3步单克隆抗体-生物素-亲和素免疫过氧化物酶技术,对8名用镍等斑贴试验阳性患者和2名漆树皮炎患者的共11份皮损活检标本进行了免疫组织学研究.结果表明,11份受检标本中的淋巴样浸润几乎均是Leu-1~+、4~+、5~+表型的成熟T细胞,其中大多数(60~90%,中位数为75%)T细胞为Leu-9~+,67~99%(中位数为95%)是Leu-3~+的辅助/诱导性T细胞亚群,平均只有5%的T细胞是Leu-2~+的细胞毒/抑制性T细胞亚群.每一标本中均含有Leu-3~+、8~+和Leu-3~+8~-T细胞,仅一份对羟化香茅醛斑贴试验阳性的标本含很少量的Leu-3~+8~+亚群.这些结果提示,与体外试验相似,Leu-3~+T细胞中的Leu-8~+和Leu-8~-亚群均参与体细内细胞介导的免疫
Recently, studies using allogeneic mixed leukocyte reactions have shown that Leu-8 + and Leu-8 ~ subsets in Leu-3 ~ + T cells play an important role in cell-mediated immune responses in vitro.In order to determine Whether they are involved in cell-mediated immune response in vivo, and confirm the expression of HLA-DR, HLA-DQ, T cell and Ki-67 antigen of many kinds of cells, Leu-3 expression of Langerhans cell, Immunohistochemistry was performed on 11 skin lesion biopsy specimens from 8 patients with positive patch tests such as nickel and 2 patients with dermatophytosis using the monoclonal antibody-biotin-avidin immunoperperoxidase technique. The results showed that almost all lymphoid infiltrates in 11 specimens tested were mature T cells of the Leu-1 +, 4 +, and 5 + phenotypes, with the majority (60-90%, median 75 %) T cells were Leu-9 ~ +, 67-99% (median 95%) were helper / inducible T cell subsets of Leu-3 ~ + with on average only 5% of T cells were Leu-2 ~ + Cytotoxic / suppressive T cell subsets.Each sample contains Leu-3 ~ +, 8 ~ + and Leu-3 ~ + 8 ~ -T cells, only one of the hydroxylated citronella aldehyde spots Test positive specimens containing a small amount of Leu-3 ~ +8 ~ + subgroups The results suggest that Leu-8 ~ + and Leu-8 ~ - subpopulations in Leu-3 ~ + T cells are all involved in the process of in vivo cell-mediated immunity
