目的 研究叶酸(FA)影响白血病细胞增殖和细胞周期分布,以探讨FA 抑制白血病细胞增殖的机理。方法 体外培养白血病细胞株HL-60、K562。利用台盼蓝拒染法进行细胞计数,流式细胞术检测细胞周期分布。结果 10 nm ol/L和300 nm ol/L的FA对HL-60细胞无显著性抑制。而100 nm ol/LFA对HL-60 细胞作用48 h 后出现显著性抑制作用。不同的是,10 nm ol/L和100 nm ol/LFA对562 细胞具有显著性促进作用。FCM 结果提示:100 nm ol/LFA 对HL-60 细胞的抑制是由于G1 期阻滞所致,而对于K562 细胞则具有促进增殖生长的作用。结论FA 的作用机制可能类似于Vit B12,具有促癌和抗癌的双重作用。我们的结果提示不能单纯地给白血病患者补充FA,而应与其它抗癌药物结合共同使用,以达到最佳治疗效果
Objective To study the effect of folic acid (FA) on the proliferation and cell cycle distribution of leukemia cells in order to investigate the mechanism of FA inhibition on proliferation of leukemia cells. Methods The leukemic cell lines HL-60 and K562 were cultured in vitro. The cells were counted using trypan blue dye exclusion and flow cytometry was used to detect cell cycle distribution. Results There was no significant inhibition of HL-60 cells by 10 nmol/L and 300 nmol/L FA. However, 100 nm ol/LFA significantly inhibited HL-60 cells for 48 h. The difference is that 10 nm ol/L and 100 nm ol/LFA have a significant promoting effect on 562 cells. FCM results suggested that inhibition of HL-60 cells by 100 nm ol/LFA was due to G1 arrest and K562 cells to promote proliferation and growth. Conclusions The mechanism of action of FA may be similar to that of Vit B12, which has a dual role of cancer promotion and anti-cancer. Our results suggest that FA can not be simply added to leukemia patients, but should be combined with other anti-cancer drugs to achieve the best therapeutic effect
