【目的】观察补肾活血方对骨关节炎软骨细胞中低氧诱导因子(HIF)-1αmRNA表达的影响。【方法】32只SD大鼠随机分为正常组、模型组、中药组、塞来昔布组(西药组),每组8只,按照Hulth法复制膝骨关节炎模型,取关节软骨,对关节软骨病理切片采用Safranin O染色,光镜下观察,进行Mankin评分,并采用实时荧光定量聚合酶链反应检测各组HIF-1αmRNA的表达。【结果】模型组Safranin O染色的Mankin评分较正常组显著升高(P<0.05);中药组Safranin O染色减少或缺失程度较西药组、模型组轻,差异有显著性意义(均P<0.05)。模型组HIF-1αmRNA表达较正常组升高,差异有显著性意义(P<0.01);中药组HIF-1αmRNA表达低于西药组、模型组,差异有显著性意义(均P<0.05)。【结论】补肾活血方可能通过抑制HIF-lαmRNA上调,从而延缓骨关节炎的发展。
【Objective】 To observe the effect of Bushen Huoxue Recipe on the expression of hypoxia inducible factor (HIF) -1α mRNA in osteoarthritic chondrocytes. 【Methods】 Thirty-two SD rats were randomly divided into normal group, model group, traditional Chinese medicine group and celecoxib group (Western medicine group), with 8 rats in each group. The knee osteoarthritis model was made according to Hulth method, The articular cartilage sections were stained with Safranin O, observed under light microscope, Mankin score, and the expression of HIF-1αmRNA in each group were detected by real-time fluorescence quantitative polymerase chain reaction. 【Results】 The Mankin score of Safranin O staining in model group was significantly higher than that in normal group (P <0.05). The decrease or deletion of Safranin O in Chinese herbal medicine group was lighter than western medicine group and model group (P <0.05) ). The expression of HIF-1αmRNA in the model group was significantly higher than that in the normal group (P <0.01). The expression of HIF-1αmRNA in the Chinese herbal medicine group was lower than that in the western medicine group and the model group (all P <0.05). 【Conclusion】 Bushen Huoxue Prescription can delay the development of osteoarthritis by inhibiting the up-regulation of HIF-1αmRNA.
