目的预测和分析肿瘤分化相关蛋白HIV-ep2/MIBP1/MBP2的结构及功能。方法检索蛋白质全序列,应用所克隆的基因片段序列Genebank数据库中进行BLUST比对,得出HIV- ep2/MIBP1的全长序列。应用ExPASy提供的ProtParam程序对该蛋白质进行基本特征进行运算,进一步进行二级结构分析。SMART预测功能域,PDB网站模拟三维结构,ELM预测基元。蛋白质亚细胞定位的信息学预测;Source资源网站作组织表达分布。结果HIV-ep2/MIBP1/MBP2基因编码一个含有2500氨基酸的巨大蛋白质,属于不稳定蛋白。二级结构主要包括62.52%随机卷曲;22.36%α螺旋;10.32%伸展链;4.80%β转角。三级结构主要含有4个锌指结构,计算机预测了蛋白的三维立体结构,可能的基元46种。功能方面该蛋白定位于细胞核,在滤泡淋巴瘤、肾近曲小管、大脑尾状核、精神分裂症脑S-11额叶、全脑、丘脑中含量丰富。结论HIV-ep2/MIBP1/MBP2蛋白为一个2500氨基酸的巨大蛋白,主要结构为4个锌指,定位于细胞核,可能与cyclin系统、Src家族等有关。
Objective To predict and analyze the structure and function of tumor-associated protein HIV-ep2 / MIBP1 / MBP2. Methods The full-length sequence of HIV-ep2 / MIBP1 was obtained by BLAST analysis using Genebank database of cloned gene sequences. Using the ProtParam program provided by ExPASy, the basic characteristics of the protein were calculated and the secondary structure was further analyzed. Results The HIV-ep2 / MIBP1 / MBP2 gene encoded a huge protein containing 2500 amino acids, belonging to the unstable protein. The secondary structure mainly includes 62.52% random crimp, 22.36% alpha helix, 10.32% extension chain and 4.80% beta angle. The tertiary structure mainly contains four zinc finger structures, and the computer predicts the three-dimensional structure of the protein and 46 possible motifs. Functional aspects of the protein located in the nucleus, in follicular lymphoma, renal proximal tubules, caudate nucleus, schizophrenia brain S-11 frontal lobe, whole brain, thalamus content. Conclusion The HIV-ep2 / MIBP1 / MBP2 protein is a 2500 amino acid macrophages whose main structure is four zinc fingers located in the nucleus, which may be related to cyclin system and Src family.
