目的　构建淡色库蚊对溴氰菊酯抗药性品系cDNA表达文库。方法　应用定向克隆技术将相应cDNA片段插入λExCell/NotⅠ /EcoRⅠ /CIP载体的NotⅠ和EcoRⅠ双酶切位点 ;并用PCR对插入片段大小进行分析。结果　获得含 4.72×10 5个重组子的cDNA表达文库 ,重组率为 10 0 % ,插入片段大小平均约 1.1kb。结论　所建文库容量及插入片段大小适合进一步研究。 Objective To construct a cDNA expression library of deltamethrin - resistant strains of Culex. Methods The corresponding cDNA fragments were inserted into the Not Ⅰ and EcoR Ⅰ restriction sites of λExCell / NotⅠ / EcoRI / CIP vector by directional cloning technique. The size of the inserted fragment was analyzed by PCR. As a result, a cDNA expression library containing 4.72 × 10 5 recombinants was obtained, with a recombination rate of 100% and an average size of the inserted fragments of about 1.1 kb. Conclusion The constructed library size and insert size are suitable for further study.
