为了精确测定柿属植物叶片的多酚含量,本研究通过单因素试验和正交试验确定Folin-ciocalteu法测定柿属植物叶片多酚含量的最佳条件,然后利用优化后的方法测定20份柿属植物叶片的多酚含量。结果表明:样品提取液中需依次加入Folin-ciocalteu试剂(0.25 M)2.5 mL,和10%Na_2CO_3溶液3 mL,再用去离子水定容至10 mL,该溶液室温显色1h后在765 nn波长处吸光度最大,且多酚在质量浓度为(2~14)ug/mL范围内与吸光度呈良好线性关系。改良后的方法具有较好的稳定性、重现性、精密度及加标回收率,其相对标准偏差分别为0.58%、0.69%、0.16%和0.96%。所测定的20份柿属植物叶片多酚含量丰富,平均含量为(44.19±8.30)mg/g,其中美洲柿含量最高,可达(80.83±0.95)mg/g。本研究一方面优化了柿属植物叶片多酚含量的Folin-ciocalteu测定法,为精确测定柿属植物叶片多酚含量提供了技术支撑;另一方面测定了具有代表性的柿属植物叶片多酚含量,对叶片资源的深入开发利用具有指导意义。
In order to determine the polyphenol content of persimmon leaves accurately, the optimal conditions of folin-ciocalteu determination of polyphenol content in leaves of persimmon were determined by single factor test and orthogonal test.
