用RAPD技术对 4类紫菜 (Porphyrayezoensis,P .haitanensis,P .katadaivar.hemiphylla和P .oligosper matangia)的 15个无性系丝状体进行了遗传多样性分析 ,从 5 0个OPERON引物中经过初筛 ,其中 6个引物可以扩增出稳定的可重复的图谱。这 6个引物共扩增出了 6 9条带 ,多态性比例达 97.1%。根据RAPD结果将这 15个无性系聚类为 3个群 ,此聚类结果与分类地位基本相符。利用来自引物OPJ_18和OPN_0 2扩增的 8条多态性条带构建了紫菜的DNA指纹图谱 ,该图谱可以方便地应用于紫菜无性系的种质鉴定。本研究还回收并克隆了 5个紫菜无性系的 5条特异片段 ,将用于无性系鉴定、种质资源保护和产权保护。
Genetic diversity analysis of fifteen clonal filaments from 4 species of seaweed (Porphyrayezoensis, P. haitanensis, P. katadaivar.hemiphylla and P.oligosper matangia) was carried out using RAPD technique, and preliminary screening was carried out from 50 OPERON primers , Of which 6 primers can amplify a stable repeatable map. A total of 69 bands were amplified with these 6 primers, and the percentage of polymorphisms was 97.1%. According to the results of RAPD, the 15 clones were clustered into three groups, and the clustering result basically accorded with the taxonomic status. The DNA fingerprinting of seaweed was constructed by using eight polymorphic bands amplified from primers OPJ_18 and OPN_02.It can be conveniently applied to the germplasm identification of laver clones. In this study, five specific fragments of five laver clones were recovered and cloned, which will be used for clonal identification, germplasm resources protection and property protection.
