目的探讨健脾益肠散对溃疡性结肠炎(UC)大鼠Toll样受体4(TLR4)的影响。方法将60只大鼠随机分为正常组、模型组、健脾益肠散高、中、低剂量组和柳氮磺吡啶组各10只;除正常组外,其余均采用2、4、6-三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型;给药21d后,观察各组大鼠给药前后的体质量变化,用酶联免疫吸附测定法、免疫组织化学法分别检测各组大鼠血清TLR4水平和结肠组织TLR4蛋白表达情况。结果模型组大鼠体质量增长明显低于正常组(P<0.01),健脾益肠散高、中剂量组能明显促进UC大鼠体质量增长(P<0.05)。模型组大鼠血清TLR4水平和结肠组织TLR4蛋白表达均明显高于正常组(P<0.01);健脾益肠散高剂量组TLR4水平和高、中剂量组TLR4蛋白表达均较模型组降低,差异有统计学意义(P<0.05或P<0.01);健脾益肠散高剂量组对TLR4的影响与柳氮磺吡啶组比较差异不显著(P>0.05),但明显优于低剂量组(P<0.05)。结论健脾益肠散对UC肠粘膜免疫的保护作用可能与降低TLR4水平及其蛋白表达有关。
Objective To investigate the effects of Jianpi Yiganchang on Toll-like Receptor 4 (TLR4) in rats with ulcerative colitis. Methods Sixty rats were randomly divided into normal group, model group, spleen and intestine scattered high, medium and low dose group and sulfasalazine group of 10; except the normal group, the rest were used 2,4,6 - trinitrobenzene sulfonic acid (TNBS) / ethanol method of UC rat model; 21d after administration, before and after administration to observe the changes in body weight in rats, using enzyme-linked immunosorbent assay, immunohistochemistry Serum levels of TLR4 and TLR4 protein in colonic tissue were detected. Results The body weight of rats in model group was significantly lower than that in normal rats (P <0.01). The spleen and intestine of rats in high and medium dose groups could obviously promote the body weight of UC rats (P <0.05). TLR4 level and TLR4 protein expression in colon tissue of model group were significantly higher than those in normal group (P <0.01). TLR4 level in high-dose Jianpi Yiganchang group and TLR4 protein in high-dose and middle-dose group were lower than those in model group (P <0.05 or P <0.01). The effect of Jianpianyicosan high dose group on TLR4 was not significantly different from that of sulfasalazine group (P> 0.05), but it was obviously better than that of low dose group (P <0.05). Conclusion Jianpi Yigan San protective effect on UC intestinal mucosa may be related to the decrease of TLR4 level and its protein expression.
