目的:为了检测细胞间隙连接蛋白基因家族中哪些成员在鼻咽癌细胞中表达,研究分化诱导剂对这些基因的表达作用和机制。方法:细胞生长曲线,Northern杂交。结果:细胞间隙连接蛋白基因(connexin,Cx)家族成员的两个基因:Cx37、Cx46在鼻咽癌细胞株HNE1表达。维甲酸能抑制HNE1的生长和Cx37和Cx46在HNE1中的表达,并能诱导RARa的表达。去除维甲酸处理后细胞生长抑制被解除,Cx37、Cx46、RARa的表达恢复正常。结论:Cx37和Cx46是鼻咽癌细胞中特异表达的Cx家族基因,维甲酸能抑制Cx46和Cx37的表达,可能的机制是通过诱导RARa的表达而起作用,维甲酸对Cx基因表达调节是可逆的。
OBJECTIVE: To investigate which members of the cell gap junction protein gene family are expressed in nasopharyngeal carcinoma cells and to investigate the role of differentiation inducing agents in the expression of these genes. Methods: Cell growth curve, Northern blotting. Results: Two genes of Cx family member: Cx37 and Cx46 were expressed in nasopharyngeal carcinoma cell line HNE1. Retinoic acid inhibits the growth of HNE1 and the expression of Cx37 and Cx46 in HNE1 and induces RARa expression. The cell growth inhibition was relieved after removing the retinoic acid, and the expression of Cx37, Cx46 and RARa returned to normal. CONCLUSION: Cx37 and Cx46 are specific Cx family genes in nasopharyngeal carcinoma cells. Retinoic acid can inhibit the expression of Cx46 and Cx37. The possible mechanism may be through the induction of RARa expression. The retinoic acid regulation of Cx gene expression is reversible of.
