目的:检测贲门腺癌(gastric cardia adenocarcinoma,GCA)组织中长链非编码RNA(long non-colding RNA,lncRNA)XLOC_008370的表达及其甲基化状态。方法:分别应用RT-PCR以及甲基化特异性PCR(methylationspecific PCR,MSP)法检测76例GCA组织及其相应的癌旁组织中XLOC_008370的表达及其甲基化状态。结果:GCA组织中XLOC_008370的表达水平显著低于其相应的癌旁组织(P<0.05),并且与GCA患者的TNM分期和淋巴结转移有关(P<0.05和P<0.01)。GCA组织中XLOC_008370启动子区的甲基化率显著高于相应的癌旁组织(P<0.05),并且与GCA患者的TNM分期和淋巴结转移有关(P<0.01和P<0.05)。发生XLOC_008370甲基化的GCA组织中XLOC_008370的表达水平显著低于未发生甲基化的GCA组织(P<0.05)。结论:GCA组织中XLOC_008370低表达,其启动子区甲基化可能是导致其表达沉默的机制之一。
OBJECTIVE: To detect the expression and methylation status of long non-coloding RNA (lncRNA) XLOC_008370 in gastric cardia adenocarcinoma (GCA). Methods: The expression and methylation status of XLOC_008370 in 76 GCA tissues and corresponding paracancerous tissues were detected by RT-PCR and methylationspecific PCR (MSP). Results: The expression level of XLOC_008370 in GCA tissues was significantly lower than that in the corresponding paracancerous tissues (P <0.05), and correlated with TNM stage and lymph node metastasis (P <0.05 and P <0.01). The methylation rate of XLOC_008370 promoter region in GCA tissues was significantly higher than that in corresponding adjacent tissues (P <0.05), and was related to TNM stage and lymph node metastasis (P <0.01 and P <0.05). The expression level of XLOC_008370 in XLOC_008370 methylated GCA tissues was significantly lower than that in non-methylated GCA tissues (P <0.05). Conclusion: The low expression of XLOC_008370 in GCA tissues may be one of the mechanisms leading to its silent expression.
