目的:建立随机案件海洛因滥用人员尿液的代谢组学研究模型,研究海洛因滥用与人体内生物标记物特异性变化关系在研究海洛因依赖的潜在价值。方法:随机选取日常案件中符合年龄、疾病史、性别比等要求,且经体金单克隆尿检板初筛、气质联用仪(GC/MS)确证滥用过海洛因的尿液30份,同时选取30例健康志愿者尿液,并用UPLC-MS分析尿液,经Mzmine 2.1软件峰匹配和多种统计学分析,筛选出有差异性变化的海洛因滥用生物标记物。再收集6份尿液重复实验对结果进行验证。结果:健康志愿者尿液与首次实验和验证实验的海洛因滥用者尿液可以实现分离,后两者存在交叉现象,说明健康组与滥用组尿液中生物标记物间存在特异性差异。结论:海洛因滥用形成依赖使得尿液中生物标记物发生了变化,经统计学初步筛选出90个特异性的生物标记物,说明借助代谢组学技术研究尿液中生物标记物的特异性变化来推断机体是否对海洛因依赖具有一定的可行性。
OBJECTIVE: To establish a metabolomics study model of urinary excretion of heroin abusers in randomized cases and investigate the relationship between the abuse of heroin and the specific changes of human biomarkers in the study of the potential value of heroin dependence. Methods: Randomly selected 30 cases of urine which were consistent with the requirements of age, disease history, sex ratio and so on, and were confirmed by GC / MS and GC / MS. Urine was obtained from 30 healthy volunteers. Urine was analyzed by UPLC-MS. Mismatched heroin abuse biomarkers were screened by Mzmine 2.1 software peak matching and various statistical analyzes. Another 6 urine samples were collected and the results were validated. Results: The urine of healthy volunteers could be separated from the urine of heroin abusers in the first experiment and verification experiment. There was a cross between the urine and healthy volunteers, indicating that there was a specific difference between the biomarkers in the urine of the healthy group and the abuse group. CONCLUSIONS: The dependence of heroin abuse leads to the change of biomarkers in urine. Ninety-nine specific biomarkers were initially screened by statistics, indicating that the specific changes of biomarkers in urine can be studied by means of metabonomics To infer whether the body has a certain feasibility of heroin dependence.
