目的探讨雌激素对兔腹主动脉球囊损伤反应的影响。方法日本雌性和雄性大耳白兔60只,随机分为假手术组、对照组、干预组,每组雌、雄各10只。假手术组行股动脉切开术,另两组以球囊导管损伤兔腹主动脉内膜,制作球囊损伤模型,干预组每天肌注17-β雌二醇(17-βE_2、100μg/kg),另两组每天肌注300μl棉子油。2周后,测定NO和内皮素(ET-1)后;动物处死取腹主动脉标本,作病理学切片,HE染色,测定内膜面积、中膜面积、内膜面积/中膜面积比值;免疫组织化学染色增殖细胞核抗原,并计算其增殖指数(PI)。结果与假手术组比较,干预组雄性兔和对照组雄、雌性免血管壁增厚、管腔变小,NO含量下降(P<0.01),ET-1含量上升(P<0.01),干预组雌性兔变化不显著(P>0.05)。干预组雄性兔和对照组雄、雌性兔ET-1、内膜面积、中膜面积、内膜面积/中膜面积比值、PI均较假手术组增加(P<0.01),而干预组雌性兔ET-1较假手术组无显著变化(P>0.05)。内膜面积、中膜面积、内膜面积/中膜面积比值、PI虽较假手术组增加(P<0.01),但较对照组明显减少(P<0.01)。结论雌激素对雄性兔血管球囊损伤无干预作用,对雌性兔血管球囊损伤有保护作用。
Objective To investigate the effect of estrogen on the injury of abdominal aorta balloon in rabbits. Methods Sixty Japanese female and male white rabbits were randomly divided into sham operation group, control group and intervention group, with 10 female and male in each group. Sham-operation group underwent femoral artery dissection. The other two groups were injured by balloon catheter intima-aortic intima of rabbits to make balloon injury model. The intervention group was intramuscularly injected with 17-β estradiol (17-βE 2, 100 μg / kg ), The other two groups daily intramuscular injection of 300μl cottonseed oil. After 2 weeks, NO and ET-1 were determined. The animals were sacrificed and the abdominal aorta specimens were taken for histopathological examination. HE staining was performed to determine the intima, intima, intima / Immunohistochemistry staining for proliferating cell nuclear antigen, and calculate the proliferation index (PI). Results Compared with the sham operation group, the male and female rabbits in the intervention group were thicker than the male and female, the lumen was smaller, the content of NO was decreased (P <0.01) and the content of ET-1 was increased (P <0.01) Female rabbits did not change significantly (P> 0.05). ET-1, intimal area, medial area, intimal area / medial area ratio and PI in male and female rabbits in intervention group were significantly higher than those in sham operation group (P <0.01) ET-1 than the sham operation group no significant change (P> 0.05). Intimal area, medial area, intimal area / medial area ratio, PI were significantly higher than those in sham operation group (P <0.01), but were significantly lower than those in control group (P <0.01). Conclusion Estrogen has no effect on the balloon injury in male rabbits and protects the female rabbits from balloon injury.
