目的:考察超声波法提取高山红景天超微粉中红景天苷和苷元的方法,并建立测定高山红景天超微粉中其含量的HPLC方法。方法:采用全自动激光粒度测试仪测定高山红景天超微粉的粒径,采用超声波法提取高山红景天超微粉中红景天苷和苷元,并采用HPLC分析法测定其含量。结果:红景天苷与红景天苷元的质量浓度在1~50 mg.L-1范围内与峰面积呈良好的线性关系,其线性方程分别为Y=2 793.7C–2 340.3(r=0.999 8)、Y=6 460.7 C-1 072.2(r=0.999 9)。当信噪比S/N≥3时,红景天苷与红景天苷元的最低检测质量浓度分别为0.25,0.12 mg.L-1。高山红景天超微粉的平均粒径为(46.0±3.6)μm;超声时间为60min时红景天苷和苷元的释放量达到最高值,且红景天苷与苷元的平均百分含量分别为0.516%,0.075%。结论:所采用的HPLC分析方法简便易行、准确,适合用于高山红景超微粉中红景天苷和苷元含量测定。
OBJECTIVE: To investigate the method of extracting salidroside and aglycone in the ultrafine powder of Rhodiola sachalinensis by ultrasonic method, and to establish HPLC method for determination of its content in Rhodiola. Methods: Particle size of Rhodiola sachalinensis was determined by automatic laser particle size analyzer. Salidroside and aglycone were extracted by ultrasonic method and the content was determined by HPLC. Results: The concentration of salidroside and salidroside in the range of 1 ~ 50 mg.L-1 showed a good linear relationship with the peak area, the linear equations were Y = 2 793.7C-2 340.3 (r = 0.999 8), Y = 6 460.7 C-1 072.2 (r = 0.999 9). When the signal to noise ratio S / N≥3, the minimum detectable concentrations of salidroside and salidroside were 0.25, 0.12 mg.L-1, respectively. The mean particle size of Rhodiola rosea was (46.0 ± 3.6) μm. The release of salidroside and aglycone reached the highest value at 60 min, and the average content of salidroside and aglycone Respectively 0.516% and 0.075%. Conclusion: The HPLC method is simple, accurate and suitable for the determination of salidroside and aglycone in the powder of Alpine Rhodiola.
