目的检测凋亡抑制蛋白PED/PEA-15在胃癌中的表达,以探讨其与胃癌发生的联系及临床意义。方法收集山西医科大学第一附属医院2012年1月至12月间行手术切除且病理证实的胃癌手术标本,应用免疫组化方法分别检测40例胃癌组织、40例相应癌旁组织及40例相应肿瘤旁正常组织中凋亡抑制蛋白PED/PEA-15的表达情况,实验结果根据阳性细胞的百分率和染色强度进行评价,三组间样本率的比较用行×列无序列联表χ2检验(检验水准α=0.05),两组间样本率的比较用独立样本的χ2检验(检验水准α=0.0167),PED/PEA-15表达与胃癌临床病理关系分析采用四格表资料χ2检验(检验水准α=0.05)。结果凋亡抑制蛋白PED/PEA-15在胃癌组织中的阳性率为70%(28/40),在癌旁组织和正常组织中阳性率分别为25%(10/40)、13%(5/40),胃癌组织中PED/PEA-15表达较癌旁组织、肿瘤旁正常组织显著增高(P<0.05)。PED/PEA-15的表达与肿瘤细胞分化和临床分期相关(P<0.05),与不同性别、年龄、肿瘤大小、转移无关(P均>0.05)。结论 PED/PEA-15在胃癌组织中高表达,可能在胃癌发生发展过程中起着重要的作用,可作为判断人胃癌生物学行为的参考指标,且有望成为胃癌靶向基因治疗的新靶点。
Objective To detect the expression of PED / PEA-15, a protein of apoptosis, in gastric cancer and to explore the relationship between PED / PEA-15 and gastric cancer and its clinical significance. Methods Surgical resection and pathological examination of gastric cancer specimens collected from January to December 2012 in the First Affiliated Hospital of Shanxi Medical University were collected. Forty gastric cancer tissues, 40 corresponding paracancerous tissues and 40 corresponding gastric cancer tissues were detected by immunohistochemistry The expression of PED / PEA-15, a protein of apoptosis in normal tissues, was evaluated according to the percentage of positive cells and the intensity of staining. The comparison of the sample rates among three groups was performed using rows × columns with unwired chi-square test (test (Α = 0.0167). The relationship between the expression of PED / PEA-15 and the clinicopathological features of gastric cancer was analyzed using the chi square test (χ2 test) = 0.05). Results The positive rate of PED / PEA-15 was 70% (28/40) in gastric cancer tissues and 25% (10/40) in paracancerous tissues and normal tissues respectively. The positive rate of PED / PEA-15 was 13% / 40). The expression of PED / PEA-15 in gastric cancer tissues was significantly higher than that in paracancerous tissues and adjacent normal tissues (P <0.05). PED / PEA-15 expression was correlated with tumor cell differentiation and clinical stage (P <0.05), but not with gender, age, tumor size and metastasis (P> 0.05). Conclusion The high expression of PED / PEA-15 in gastric cancer may play an important role in the development and progression of gastric cancer. It may serve as a reference index for judging the biological behavior of human gastric cancer and is expected to become a new target of targeted gene therapy for gastric cancer.
