目的:研究罗汉果悬浮细胞生长、罗汉果甜苷积累的动力学特征,为培养过程的优化控制提供理论依据。方法:考察1个培养周期内罗汉果悬浮细胞的鲜量重、干质量和罗汉果甜苷的含量,绘制生长曲线和生长速率曲线。结果:罗汉果悬浮细胞培养的生长周期约为21 d,0~6 d为细胞的延滞期,7~21 d为对数期,稳定期较短,第22天进入到衰亡期,在第21天时细胞鲜、干质量达到最大,分别为693.0,416.0 g.L-1,最大生长速率为0.040 g.L-1.d-1。罗汉果总苷和甜苷V从第7天开始快速积累,第21天达到最大值,生长速率分别为0.032 8,0.054 7 g.L-1.d-1。结论:联合研究细胞生长和产物积累,可以明确产物积累的动态变化规律,为培养过程的优化、代谢产物的调控和大规模的工业化生产提供实验依据。
OBJECTIVE: To study the kinetic characteristics of the growth of suspension cells and the accumulation of mogrosides in Lo Han Guo and provide a theoretical basis for the optimal control of the culture process. Methods: The fresh weight, dry weight and the content of mogroside in Lo Han Guo suspension cells were investigated in one culture cycle, and the growth curve and growth curve were drawn. Results: The growth period of Lo Han Guo suspension cells culture was about 21 days. The lag phase was 0-6 days, and the logarithmic phase was from 7 to 21 days. The stability period was short and reached the decay stage on the 22nd day. On the 21st day, The fresh and dry weight of cells reached 693.0 and 416.0 gL-1, respectively. The maximum growth rate was 0.040 gL-1.d-1. Mogroside and glycoside V accumulated quickly from the 7th day and reached the maximum on the 21st day with the growth rates of 0.032 8 and 0.054 7 g.L-1.d-1, respectively. Conclusion: The combination of cell growth and product accumulation can clarify the dynamic changes of product accumulation and provide experimental basis for the optimization of culture process, regulation of metabolites and large-scale industrial production.
