The development of a malaria control program and its effectiveness assessment depend on detailed epidemiological data. However, the accuracy of the data obtained by microscopic examination is affected by many factors; it is also not reliable to identify the malaria parasite species according to the morphological characteristics of the malaria parasite. To this end the authors used PCR detection and identification of human and Anopheles in vivo parasite, and the microscopic examination results were evaluated. In October 1992 (dry season) and November (rainy season) in Bor village, Bissau, Guinea, 110 and 79 villagers were enrolled each to collect approximately 10 Oμl of finger-tip blood and blood smear was prepared for each person at the same time. In the second blood sampling Anopheles 75 were caught in a given room in the same village. Phenol / ethanol extraction of DNA samples carried out
