目的:研究PTHrP、Sox9和Bcl-2基因在人体软骨肉瘤细胞HTB-94中的相互作用机制。方法:小干扰siRNA阻断PTHrP后,分别检测肿瘤细胞中Sox9、Bcl-2和Col2α1的mRNA及蛋白表达。结果:PTHrP基因被干扰后,PTHrP基因的mRNA及蛋白表达降低,Sox9、Bcl-2和Col2α1的mRNA及蛋白表达都受到抑制;而PTHrP基因的干扰作用去除,Sox9、Bcl-2和Col2α1的mRNA及蛋白表达抑制作用也消失。结论:PTHrP基因在肿瘤细胞的抑制引起Sox9、Bcl-2和Col2α1肿瘤相关基因的抑制,提示PTHrP基因是Sox9、Bcl-2和Col2α1肿瘤相关基因的上游基因。
AIM: To investigate the interaction between PTHrP, Sox9 and Bcl-2 in human chondrosarcoma HTB-94 cells. Methods: Small interfering siRNAs blocked PTHrP and detected the mRNA and protein expression of Sox9, Bcl-2 and Col2α1 in tumor cells respectively. Results: The mRNA and protein expression of PTHrP gene was decreased after PTHrP gene was interfered, and the mRNA and protein expressions of Sox9, Bcl-2 and Col2α1 were inhibited. The interference of PTHrP gene was eliminated. The mRNA expressions of Sox9, Bcl-2 and Col2α1 And protein expression inhibition also disappeared. Conclusion: Inhibition of PTHrP gene in tumor cells caused the inhibition of Sox9, Bcl-2 and Col2α1 tumor-related genes, suggesting that PTHrP is an upstream gene of Sox9, Bcl-2 and Col2α1 tumor-associated genes.
