本研究利用前期研究中东方田鼠血清筛选日本血吸虫成虫噬菌体展示cDNA文库获得的10个阳性克隆中的8个噬菌体克隆单独或联合免疫BALB/c小鼠,观察对日本血吸虫病的免疫预防效果,以筛选其中具有发展成疫苗的候选抗原编码基因。研究发现与空白对照组相比,展示血吸虫锌指蛋白的1号噬菌体克隆免疫组在两次实验中分别获得了32.10%和31.25%的减虫率、61.14%和47.31%的肝脏减卵率,虫体合抱率分别由92.59%、57.39%下降到69.09%、41.03%;其它噬菌体克隆单独免疫组或联合免疫组在第一次实验中均获得了8.02%~32.72%的减虫率和40.19%~69.53%的减卵率,但在第二次实验中未能得到验证。研究结果提示,日本血吸虫环状锌指蛋白(1号克隆)编码基因在疫苗方面具有重要研究价值。
In this study, BALB / c mice were immunized with 8 phage clones of 10 positive clones obtained from phage display cDNA library of Schistosoma japonicum by serum screening of voles of Microtus fortis in pre-study to observe the immunological preventive effect on schistosomiasis japonica Candidate antigen-encoding genes were screened for development of the vaccine. The results showed that compared with the blank control group, the immunized group of phage clone 1 displaying the zinc finger protein of schistosomes obtained the worm reduction rates of 32.10% and 31.25%, the egg reduction rates of 61.14% and 47.31% respectively in two experiments, The worm encroachment rate decreased from 92.59% and 57.39% to 69.09% and 41.03%, respectively. In the first experiment, the phage clone alone or combined immunization group obtained the worm reduction rate of 8.02% -32.72% and the rate of 40.19% ~ 69.53% of the egg reduction rate, but in the second experiment failed to be verified. The results suggest that the gene encoding Schistosoma japonicum zinc finger protein (clone No. 1) has important research value in the field of vaccines.
