目的探讨RNA干扰(RNAi)Survivin基因对食管鳞癌KYSE150细胞生物学的影响。方法体外构建Survivin基因的RNA干扰(RNAi)慢病毒载体,侵染KYSE150细胞,检测Survivin基因及Caspase-3的mRNA和蛋白表达量,转染前后KYSE150细胞的增殖,凋亡变化情况。构建食管癌裸鼠移植瘤模型,观察SurvivinsiRNA对肿瘤细胞生长的影响。结果 SurvivinsiRNA抑制了KYSE150细胞的生长,降低Survivin mRNA及蛋白的表达,并诱导胞凋亡。食管癌裸鼠移植瘤转染组肿瘤较空白和阴性对照组生长明显缓慢。结论 Survivin在食管癌细胞的增殖和凋亡方面发挥着重要的调控作用。
Objective To investigate the effect of survivin gene RNAi on the biology of esophageal squamous cell carcinoma cell line KYSE150. Methods RNA interference (RNAi) lentiviral vector containing Survivin gene was constructed in vitro and infected KYSE150 cells. The mRNA and protein expression of Survivin gene and Caspase-3 were detected, and the proliferation and apoptosis of KYSE150 cells were detected. To construct esophageal carcinoma xenograft model in nude mice and observe the effect of SurvivinsiRNA on tumor cell growth. Results SurvivinsiRNA inhibited the growth of KYSE150 cells, decreased the expression of Survivin mRNA and protein, and induced apoptosis. The tumor growth in transplanted esophageal xenografts in nude mice was obviously slower than that in blank and negative control groups. Conclusion Survivin plays an important regulatory role in the proliferation and apoptosis of esophageal cancer cells.
