甘遂半夏汤对大鼠肝功能及形态学的影响

目的:探讨甘遂半夏汤水煎液对正常大鼠肝脏的慢性毒性作用。方法:将80只SD大鼠随机分为甘遂半夏汤高、中、低剂量组(37,18.5,9.25 g·kg-1)及正常组,每组20只,各组大鼠采用等体积不等浓度ig,ig体积均为0.02 m L·g-1。实验组ig给药,正常组ig给予蒸馏水。给药90 d。最后1次给药后禁食15 h,各组随机抽取一半大鼠,取血,采用全自动生化分析仪检测肝功能指标丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST),AST/ALT,总胆红素(TBIL)和直接胆红素(DBIL),及血脂指标胆固醇(CHO),甘油三酯(TG),高密度脂蛋白(HDL),低密度脂蛋白(LDL);取肝,HE染色观察大体形态和组织学形态。剩余动物停药后继续观察2周,重复以上检查指标。结果:给药期,与正常组比较,甘遂半夏汤高剂量组ALT,AST,AST/ALT,TBIL和DBIL均明显升高(P<0.01),中剂量组ALT,AST,DBIL(P<0.05)和AST/ALT均明显升高(P<0.01),低剂量组无显著变化;与正常组比较,各剂量组CHO,TG,HDL,LDL无显著性变化;随着剂量增大,肝脏组织病理变化逐渐加重。恢复期,各组大鼠各项检测指标及肝脏组织学形态无显著差异。结论:长期服用甘遂半夏汤可导致大鼠肝损伤,对大鼠肝功能及肝组织形态学的影响存在着剂量依赖性,小剂量长期使用影响较小,随着剂量的增加,影响的程度逐渐加重,但损伤具有可逆性。 Objective: To investigate the chronic toxicity of Gan Sui Banxia Decoction on normal rat liver. Methods: Eighty SD rats were randomly divided into high, medium and low doses of Gan Sui Banxia Decoction (37,18.5,9.25 g · kg-1) and normal group, with 20 rats in each group. The volumetric ig and ig volumes were 0.02 m L · g-1. Experimental group ig administration, normal group ig given distilled water. Dose 90 d. The rats were fasted for 15 h after the last administration. Half of the rats in each group were randomly selected for blood. Blood samples were collected for the detection of liver function indexes such as ALT and aspartate aminotransferase AST, AST / TB, TBIL and DBIL, as well as cholesterol, triglyceride, high density lipoprotein (HDL), low density lipoprotein (LDL); take the liver, HE staining observed gross morphology and histological morphology. The remaining animals continue to observe the withdrawal after 2 weeks, repeat the above indicators. Results: Compared with the normal group, the levels of ALT, AST, AST / ALT, TBIL and DBIL in the Kansui Banxia Decoction high dose group were significantly increased (P <0.01) 0.05) and AST / ALT were significantly increased (P <0.01), but there was no significant change in low dose group. Compared with normal group, there was no significant change of CHO, TG, HDL and LDL in each dose group; Liver tissue pathological changes gradually increased. During the recovery phase, there was no significant difference between the detection indexes and the histological changes of the liver in each group. CONCLUSION: Long-term administration of Kansui Banxia Decoction can cause liver injury in rats, and has a dose-dependent effect on liver function and liver morphology in rats. However, long-term use of low dose has little effect. With the increase of dose, Degree gradually increased, but the damage is reversible.
