优化大黄游离总蒽醌的提取工艺,为其在农业、医药、食品等方面的开发提供理论参数。在单因素试验基础上,选取提取时间、溶剂配比和液料比3个因素进行Box-Benhnken响应面法试验设计,对其提取工艺参数进行优化。在提取温度60℃,提取次数2次,提取时间2.11 h,溶剂配比0.43,液料比23.23时,大黄游离总蒽醌的理论提取率最高,可达1.021%。根据实际试验情况,将其参数修正为提取时间2.1 h,溶剂配比0.4,液料比23.2,提取温度60℃,提取次数2次,经验证此条件下大黄游离总蒽醌含量为1.017%,与理论值较为接近。采用Box-Behnken响应面法建立的游离总蒽醌提取工艺模型提取率高,并能很好地预测试验结果。
Optimize the extraction process of rhubarb free anthraquinone, which provides theoretical parameters for the development of agriculture, medicine, food and so on. Based on the single-factor experiment, the Box-Benhnken response surface methodology was used to optimize the extraction process parameters based on three factors: extraction time, solvent ratio and liquid / solid ratio. The theoretical extraction rate of free anthraquinones from Rhubarb reached 1.021% with the extraction temperature of 60 ℃, the extraction times of 2 times, the extraction time of 2.11 h, the solvent ratio of 0.43 and the liquid to liquid ratio of 23.23. According to the actual test conditions, the parameters were revised to 2.1 h extraction time, 0.4 solvent ratio, 23.2 liquid-to-liquid ratio, 60 ℃ extraction temperature and 2 extraction times. The content of free anthraquinones in Rhubarb was 1.017% Close to the theoretical value. The Box-Behnken response surface method to establish the free total anthraquinone extraction process model extraction rate is high, and can well predict the test results.
