目的:旨在研究高压氧联合电针治疗对脊髓损伤大鼠髓鞘抑制相关蛋白(Nogo-A及NgR受体)的影响。方法:将84只雌性SD大鼠按照数字表法分为4组,每组21只,依次为模型组、高压氧治疗组、电针治疗组和联合治疗(电针联合高压氧)组。建模成功后,模型组不进行干预,其余各组进行相应的治疗。分别在损伤后3、7、14 d进行取材,对损伤大鼠进行BBB评分、HE染色及相关基因检测。结果:损伤后3、7、14 d,高压氧治疗组、电针治疗组和联合治疗组的BBB评分均高于模型组(n P<0.01),且联合治疗组评分均高于高压氧治疗组和电针治疗组,高压氧治疗组与电针治疗组比较差异无统计学意义。损伤后3、7、14 d高压氧治疗组、电针治疗组和联合治疗组Nogo-A mRNA表达量均低于模型组(n P<0.05);损伤后7、14 d高压氧治疗组NgR mRNA表达量(0.49±0.53、0.52±0.47)、电针治疗组NgR mRNA表达量(0.53±0.58、0.58±0.36)和联合治疗组NgR mRNA表达量(0.16±0.13、0.20±0.13)均低于模型组NgR mRNA表达量(1.33±1.26、1.03±0.30)(n P<0.05),且联合治疗组Nogo-A及NgR mRNA表达量低于高压氧治疗组和电针治疗组(n P0.05)。n 结论:高压氧联合电针治疗脊髓损伤大鼠与单一疗法相比更能抑制Nogo-A及NgR mRNA的表达,减轻细胞损伤,促进脊髓纤维束的生长和运动功能的恢复。“,”Objective:To explore the effects of hyperbaric oxygen (HBO) combined with electroacupuncture on the myelin inhibitory associated proteins (Nogo-A and NgR receptor) in rats with spinal cord injury.Methods:A total of 84 female SD rats were randomly divided into four groups, namely, model group, HBO group, electroacupuncture group, and combined treatment group. After successful modeling, the model group did not receive intervention, and the other groups were treated accordingly. Samples were collected on the 3, 7, and 14 d after the injury for evaluating the injured rats with Basso, Beattie, Bresnahan (BBB) Locomotor Rating Scale, and performing the HE staining and related gene detection on them.Results:After the injury on the 3, 7, and 14 d, the BBB scores of the HBO group, the electroacupuncture group, and the combined treatment group were significantly higher than those of the model group (n P<0.01), and the scores of the combined treatment group were higher than those of the HBO group and the electroacupuncture group, but there was no significant difference between the score of the HBO group and that of the electroacupuncture group. After the injury on the 3, 7, and 14 d, Nogo-A mRNA expression levels in the HBO group, the electroacupuncture group, and the combined treatment group were all lower than those in the model group (n P<0.05). After the injury on the 7 d and 14 d, the expression levels of NgR mRNA in the HBO group (0.49±0.53, 0.52±0.47), the electroacupuncture group (0.53±0.58, 0.58±0.36), and the combined treatment group (0.16±0.13, 0.20±0.13) were all lower than those in the model group (1.33±1.26, 1.03±0.30) (n P0.05).n Conclusion:Compared with monotherapy, the combined treatment of spinal cord injury in rats can inhibit the expressions of Nogo-A and NgR mRNA better, reduce cell injury, and promote the growth of spinal cord fiber bundle and the recovery of motor function.
