目的:观察希美宁对2型糖尿病(T2DM)模型大鼠血清中胰高血糖素样肽-1(GLP-1)、葡萄糖依赖性促胰岛素释放肽(GIP)含量及骨骼肌中磷脂酰肌醇-3-激酶(PI3KmRNA)、丝苏氨酸蛋白激酶(AktmRNA)表达的影响。方法:脂肪乳联合2次小剂量注射链脲佐菌素(STZ)复制2型糖尿病大鼠模型,将大鼠随机分为空白组、模型组、希美宁组、二甲双胍对照组,酶联免疫法(ELISA)检测各组大鼠血清中GLP-1、GIP的含量,RT-PCR法检测各组大鼠骨骼肌中PI3KmRNA、AktmRNA的表达水平。结果:与模型组相比,希美宁组、二甲双胍对照组可增加大鼠血清中GLP-1、GIP的含量(P<0.05或P<0.01),亦可显著上调骨骼肌中PI3KmRNA、AktmRNA的表达(P<0.01)。结论:希美宁可通过促进GLP-1、GIP的分泌来调节机体代谢,亦可通过改变PI3KmRNA、AktmRNA的表达改善T2DM模型大鼠的胰岛素抵抗。
Objective: To observe the effects of Ximei Ning on the content of GLP-1, GIP in skeletal muscle of type 2 diabetes mellitus (T2DM) rats and the levels of phosphatidylinositol -3-kinase (PI3K mRNA), serine protein kinase (Akt mRNA) expression. Methods: The rats were randomly divided into blank group, model group, simmeline group, metformin control group and enzyme-linked immunosorbent assay (ELISA) for the model of type 2 diabetes mellitus (T2DM) The content of GLP-1 and GIP in sera of rats in each group were detected by ELISA. The expression of PI3K mRNA and Akt mRNA in skeletal muscle were detected by RT-PCR. Results: Compared with the model group, the Ximeilin group and the metformin control group could increase the content of GLP-1 and GIP (P <0.05 or P <0.01), and also significantly upregulate the expression of PI3KmRNA and Akt mRNA in the skeletal muscle (P <0.01). Conclusion: Ximeini can regulate the body metabolism by promoting the secretion of GLP-1 and GIP. It can also improve the insulin resistance of rats with T2DM by changing the expression of PI3K and Akt mRNA.
