目的观察山腊梅茎水提物对慢性阻塞性肺病(COPD)模型大鼠炎症细胞和C-反应蛋白(CRP)、白细胞介素(IL)-8的影响。方法采用反复烟熏加内毒素气管内滴入法复制COPD大鼠模型,将60只SD大鼠随机分为五组:正常组、模型组和低、中、高剂量山脂梅组。模型组及正常组给予生理盐水灌胃,山腊梅组分别给予低、中、高剂量进行灌胃,疗程21 d。观察各组大鼠肺组织的病理学改变,测定各组支气管肺泡灌洗液白细胞计数、中性粒细胞比率及各组血清和肺组织中CRP、IL-8水平。结果模型组大鼠出现慢性支气管炎特征性病理改变明显,气管及血管周围有大量炎症细胞浸润,肺气肿明显,低、中、高剂量治疗组肺气肿和炎症浸润较模型组有明显减轻,正常组大鼠肺组织结构基本正常。模型组大鼠肺组织和血清CRP、IL-8较正常对照组显著增多,高、中、低剂量山腊梅组大鼠肺组织和血清CRP、IL-8均明显较模型组减少(P<0.05)。模型组中白细胞总数及中性粒细胞比率较各山腊梅组增加(P<0.01),各山腊梅组白细胞计数及中性粒细胞较正常组明显增多(P<0.01)。结论山腊梅茎水提物可降低COPD大鼠肺组织和血清中CRP及IL-8水平,对COPD大鼠的炎症有抑制作用。
Objective To observe the effects of Rhizoma Paridophilum water extract on inflammatory cells, C-reactive protein (CRP) and interleukin (IL) -8 in rats with chronic obstructive pulmonary disease (COPD). Methods Duplicate smoking and endotoxin intratracheal intratracheal instillation COPD rat model, 60 SD rats were randomly divided into five groups: the normal group, the model group and low, medium and high doses of the Shan-Mei group. The model group and the normal group were given normal saline gavage, the mountain Lamei group were given low, medium and high doses for intragastric administration, the treatment of 21 d. The pathological changes of lung tissue in each group were observed. The levels of leukocyte and neutrophil in bronchoalveolar lavage fluid and the levels of CRP and IL-8 in serum and lung tissue of each group were determined. Results The pathological changes of chronic bronchitis in the model group were obvious. There was a large number of infiltration of inflammatory cells in the trachea and perivascular area with obvious emphysema. Compared with the model group, the emphysema and inflammatory infiltration in the low, medium and high dose treatment groups were significantly reduced The normal lung tissue in normal rats was normal. The levels of CRP and IL-8 in lung tissue and serum in model group were significantly higher than those in normal control group. The levels of CRP and IL-8 in lung tissue and serum of rats in high, medium and low dose groups were significantly decreased (P <0.05 ). The total number of leukocytes and neutrophil ratio in model group was higher than that in each group (P <0.01). The white blood cell count and neutrophil count in each group were significantly higher than those in normal group (P <0.01). Conclusion Rhizoma Paridis extract can reduce the levels of CRP and IL-8 in the lung tissue and serum of COPD rats and inhibit the inflammation of COPD rats.
