【目的】研究来源于鼻咽癌细胞株SUNE1及EBV标准株B95 - 8细胞中两种EBVLMP1基因在真核细胞Balb/c3T3中的表达 ,为进一步研究EBVLMP1基因的核酸疫苗打基础。【方法】将两种不同来源EBVLMP1基因的真核表达质粒 ,经脂质体法转染Balb/c 3T3细胞后 ,用Westernblot、免疫组化及PCR的方法检测不同转染细胞中EBVLMP1蛋白的表达及核酸片段。【结果】PCR结果表明在两种不同来源LMP1转染细胞中均有EBVLMP1基因序列 ,并均能表达 6 3ku的蛋白质 ,经Westernblot和免疫组化结果证实均为EBVLMP1蛋白。【结论】来源于B95 8细胞及鼻咽癌细胞SUNE1的两种EBVLMP1基因均能在真核细胞Balb/c 3T3中表达。
【Objective】 To investigate the expression of EBVLMP1 gene in Balb / c3T3 eukaryotic cells from nasopharyngeal carcinoma cell line SUNE1 and EBV standard cell line B95 - 8, so as to lay the foundation for further study of nucleic acid vaccine against EBVLMP1 gene. 【Method】 The EBVLMP1 gene eukaryotic expression plasmids from two different sources were transfected into Balb / c 3T3 cells by liposome. The expression of EBVLMP1 protein in different transfected cells was detected by Western blot, immunohistochemistry and PCR And nucleic acid fragments. 【Result】 The results of PCR showed that both EBVLMP1 gene sequences were expressed in LMP1 transfected cells from two different sources, and all of them could express 63 ku proteins. The results of Western blot and immunohistochemistry showed that EBVLMP1 protein was both EBVLMP1 protein. 【Conclusion】 Both EBVLMP1 genes derived from B95 8 cells and nasopharyngeal carcinoma SUNE1 cells can be expressed in eukaryotic cells Balb / c 3T3.
