为探索日本血吸虫重组铁蛋白的疗效考核价值。表达、纯化日本血吸虫重组铁蛋白 ( r Sjferritin) ,用免疫印迹法分析该重组蛋白的免疫活性 ;用 r Sjferritin酶联免疫吸附试验检测治疗前、后不同时期慢性血吸虫病病人的特异性 Ig G及肝吸虫、肺吸虫、旋毛虫、囊虫病病人和正常人血清中的交叉抗体。结果显示 ,r Sjferritin具有较好的免疫活性 ;用其检测日本血吸虫的敏感性和特异性与虫源性天然抗原 ( AWA和 SEA)无显著性差异 ,检测治疗后 6个月、1年和 2年慢性血吸虫病病人 Ig G的阴转率分别为 31.8% ,6 3.3%和 90 .5 % ,治疗后 Ig G水平明显下降 ,治疗后 2年与正常人无显著性差异。因此 r Sjferritin可用于诊断血吸虫病 ,并可考核疗效
In order to explore the therapeutic value of Schistosoma japonicum recombinant ferritin. The recombinant protein of Schistosoma japonicum was expressed and purified, and the immunological activity of the recombinant protein was analyzed by immunoblotting. The specific Ig G and IgM of patients with chronic schistosomiasis were detected by r Sjferritin enzyme-linked immunosorbent assay (ELISA) Liver flukes, paragonimiasis, trichinellosis, cysticercosis patients and normal human serum cross-antibodies. The results showed that r Sjferritin had good immunocompetence. The sensitivity and specificity of rSjferritin to detect Schistosoma japonicum were not significantly different from those of natural parasites (AWA and SEA). Six months, one year and two The negative conversion rates of Ig G in patients with chronic schistosomiasis were 31.8%, 63.3% and 90.5%, respectively. After treatment, the levels of Ig G were significantly decreased. There was no significant difference between the two groups after 2 years of treatment. Therefore, r Sjferritin can be used to diagnose schistosomiasis, and can assess the curative effect
