运用紫外可见分光光度法建立灵芝干粉中总灵芝酸的含量测定方法。采用70%乙醇超声提取,加入5%香草醛-冰乙酸及高氯酸显色,测定吸光度。结果表明:对照品及供试品均在560nm处有最大吸收。线性相关系数r为0.9970,精密度、重复性、回收率试验的RSD值分别为1.32%、0.83%和3.90%,平均加样回收率为97.82%。该方法简便、准确,专属性强,可作为检测灵芝干粉中总灵芝酸的方法。
Determination of Total Ganoderic Acid in Ganoderma Lucidum by UV Spectrophotometry. Ultrasonic extraction with 70% ethanol, adding 5% vanillin - acetic acid and perchloric acid color, measured absorbance. The results show that: the reference substance and the test sample all have the maximum absorption at 560nm. The linear correlation coefficient r was 0.9970. The RSD values of precision, repeatability and recovery were 1.32%, 0.83% and 3.90%, respectively. The average recoveries were 97.82%. The method is simple, accurate and specific, and can be used as a method for detecting Ganoderma lucidum in Ganoderma lucidum powder.
