作者通过体外骨髓半固体和长期液体培养(LTBMC),研究了干扰素-a(IFN-a)对慢性粒细胞白血病(CML)细胞的作用。在半固体琼脂培养体系中,IFN-a能抑制正常对照和CML患者7天和10天CFU-GM,而不抑制14天及其后的CFU-GM。在LTBMC中,于培养开始只加一次IFN-a(10 ̄2U/ml,10 ̄3U/ml,10 ̄4U/ml),对非贴壁层细胞数和基质细胞层的形成均无明显影响,但大剂量IFN-a(10 ̄3U/ml和10 ̄4U/ml组)对非贴壁层的CFU-GM抑制明显;每周连续加用IFN-a(10U/ml,10 ̄2U/ml,10 ̄3U/ml),对非贴壁层细胞数和CFU-GM均有抑制作用,而且IFN-a浓度愈高,贴壁层的形成愈少。上述结果表明:IFN-a优先抑制晚期CFU-GM,对基质细胞也有一定抑制作用。
The authors investigated the effect of interferon-a (IFN-a) on chronic myeloid leukemia (CML) cells by in vitro bone marrow semi-solid and long-term liquid culture (LTBMC). In a semi-solid agar culture system, IFN-α inhibited CFU-GM at 7 days and 10 days in normal controls and CML patients without inhibiting CFU-GM at day 14 and thereafter. In LTBMC, IFN-α (10 ~ 2U / ml, 10 ~ 3U / ml, 10 ~ 4U / ml) added at the beginning of culture showed no significant effect on the number of non-adherent cells and the formation of stromal cells (10 ~ 3U / ml and 10 ~ 4U / ml group) on the non-adherent layer of CFU-GM significantly inhibited; weekly continuous addition of IFN-a (10U / ml, ml, 10 ~ 3U / ml) on the number of non-adherent cells and CFU-GM were inhibited, and the higher the concentration of IFN-a, the formation of adherent layer less. The above results show that: IFN-a preferentially inhibits late CFU-GM, also has some inhibition of stromal cells.
