目的:通过实验观察速效平喘合剂与激素单独使用对气道重建的病理形态学改变以及对基质金属蛋白酶-9(MMP-9)的表达的影响。方法:复制大鼠哮喘模型。健康Wistar大鼠60只随机分成空白组、模型组、中药组、地塞米松组,每组15只。第1次诱喘后24小时开始给药,共治疗14天。观察速效平喘合剂与激素单独应用对气道壁MMP-9的表达的影响,并采用图像分析光密度进行评定。结果:速效平喘合剂能显著降低哮喘大鼠气道壁MMP-9的表达,干预哮喘气道重建。速效平喘合剂能显著降低大鼠哮喘模型肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)的含量,抑制肺局部EOS的聚集,明显减轻由EOS浸润引起的慢性变态性炎症反应。结论:嗜酸性粒细胞的浸润、MMP-9的表达在气道重建中发挥着重要的作用;在气道重建形成早期即应用速效平喘合剂预防性给药,可明显预防或延缓气道重建的发生。
OBJECTIVE: To observe the pathomorphological changes of airway remodeling and the effect on the expression of matrix metalloproteinase-9 (MMP-9) through the experiments of rapid-acting asthma mixture and hormone alone. Methods: Rat model of asthma was duplicated. Sixty healthy Wistar rats were randomly divided into blank group, model group, traditional Chinese medicine group and dexamethasone group, 15 rats in each group. The first 24 hours after the start of induction of drug administration, a total of 14 days. To observe the effect of quick-acting asthma mixture and hormone alone on the expression of MMP-9 in the airway wall, and evaluate the optical density by image analysis. Result: SLJF can significantly reduce the expression of MMP-9 in the airway wall of asthmatic rats, and interfere the airway reconstruction of asthma. SLJT can significantly reduce the content of eosinophils (EOS) in bronchoalveolar lavage fluid (BALF), inhibit the aggregation of local EOS in lung and significantly reduce chronic pernicious inflammatory reaction caused by EOS infiltration. Conclusion: Eosinophil infiltration and MMP-9 expression play an important role in airway remodeling. In the early stage of airway remodeling, rapid-acting anti-asthmatic agent can be used prophylactically to prevent or delay airway remodeling happened.
