目的 了解CTLA4Ig腺病毒在不同哺乳动物体内的转染能力及表达情况。方法 用所构建的CTLA4Ig腺病毒通过静脉或腹腔注射到Balb/c小鼠、Wistar大鼠、兔、小香猪、羊体内 ,应用蛋白质斑点印记检测CTLA4Ig在以上动物血清中的表达情况 ,同时还观察各动物脏器的病理改变及动物的生存情况。结果 重组腺病毒转染 7d后大鼠、小鼠血清中出现CTLA4Ig ,持续表达 2周。各动物未见异常死亡 ,各脏器未见明显病理改变。结论 CTLA4Ig重组腺病毒能够有效转染Balb/c小鼠、Wistar大鼠
Objective To understand the transfection ability and expression of CTLA4Ig adenovirus in different mammals. Methods The constructed CTLA4Ig adenovirus was injected into Balb / c mice, Wistar rats, rabbits, small pigs and sheep by intravenous or intraperitoneal injection. The expression of CTLA4Ig in serum of the above animals was detected by protein blotting. Observe the pathological changes of various animal organs and animal survival. Results After 7 days of transfection, the CTLA4Ig appeared in the serum of mice, which was continuously expressed for 2 weeks. No abnormal deaths of animals, no obvious pathological changes in various organs. Conclusion CTLA4Ig recombinant adenovirus can effectively transfect Balb / c mice, Wistar rats
