【目的】研究双歧杆菌对坏死性小肠结肠炎(necrotizing enterocolitis,NEC)模型新生SD大鼠肠组织TNF-α及IL-10水平的影响,探讨双歧杆菌对NEC的保护作用。【方法】2日龄SD大鼠128只随机分成LPS组(A组)、LPS+双歧杆菌组(B组)、生理盐水对照组(C组)、双歧杆菌对照组(D组),一周后,腹腔注射LPS后2、6、12、24h断头处死大鼠,解剖留取肠管,其中回盲部近端肠管行病理学检查,剩余肠管ELISA法检测TNF-α、IL-10水平。【结果】B组2、6、12、24h肠组织病理损伤评分均显著低于A组(P<0.05)。B组2、6、12肠组织TNF-α水平均显著低于A组(P<0.05);B组2、6、12、24肠组织IL-10水平均显著高于A组(P<0.05)。统计学表明:肠组织病理损伤评分与TNF-α水平呈正相关性(r=0.646),肠组织病理损伤评分与IL-10水平呈负相关性(r=-0.598)。【结论】双歧杆菌能抑制NEC模型中TNF-α水平和提升IL-10水平,对NEC具有保护作用。
【Objective】 To investigate the effect of Bifidobacterium on TNF-α and IL-10 in intestinal tissue of neonatal SD rats with necrotizing enterocolitis (NEC) model and to explore the protective effect of Bifidobacterium on NEC. 【Methods】 One hundred and eighty two-day-old SD rats were randomly divided into four groups: LPS group (A group), LPS + Bifidobacterium group (B group), saline control group (C group), Bifidobacterium control group (D group) After intraperitoneal injection of LPS, rats were sacrificed at 2, 6, 12, and 24 hours after the LPS injection. The intestine was dissected and the proximal intestine of the ileocecal region was examined by pathology. The levels of TNF-α and IL-10 were detected by ELISA. 【Results】 The pathological damage scores of intestinal tissue in group B at 2, 6, 12 and 24 hours were significantly lower than those in group A (P <0.05). The levels of IL-10 in 2,6,12,24 intestinal tissue in group B were significantly higher than those in group A (P <0.05) ). The statistics showed that there was a positive correlation between pathological damage score of intestinal tissue and TNF-α level (r = 0.646), negative correlation between pathological damage score of intestinal tissue and IL-10 level (r = -0.598). 【Conclusion】 Bifidobacterium can inhibit the level of TNF-α and increase the level of IL-10 in NEC model, which has a protective effect on NEC.
