[目的]研究锰对大鼠生精细胞caspase-3表达的影响,探讨锰对大鼠生精细胞caspase-3表达作用的机理。[方法]雄性SD大鼠(体重120±10g)24只随机分为3组:空白对照组,15mg/kg和30mg/kgMnCl2组。8只/组。MnCl2组分别染锰(MnCl2·4H2O)4w,空白对照组给予等容NS,给药途径均为腹腔注射,5d/w,1次/d,于第4w末处死,免疫组化(SABC法)法检测睾丸caspase-3和cyto-c表达。[结果]与空白对照组比较,各染锰组cas-pase-3阳性细胞率均显著升高,cyto-c阳性细胞率均显著降低(P﹤0.01)。染锰时间相同,30mg/kgMnCl2组与15mg/kgMnCl2组比较,caspase-3阳性细胞率显著升高,cyto-c阳性细胞率显著降低(P﹤0.01)。各组caspase-3和cyto-c阳性细胞率呈显著负相关(r=-0.876,P﹤0.01)。[结论]锰可通过线粒体途径诱发大鼠生精细胞凋亡,且caspase-3的表达随染锰剂量的增加而增加,二者存在一定的剂量-效应关系。
[Objective] To investigate the effect of manganese on the expression of caspase-3 in rat spermatogenic cells and the mechanism of manganese on the expression of caspase-3 in rat spermatogenic cells. [Methods] Twenty-four male Sprague-Dawley rats weighing 120 ± 10g were randomly divided into three groups: blank control group, 15mg / kg and 30mg / kg MnCl2 group. 8 / group. The routes of administration were intraperitoneal injection, 5 d / w, once per day, and were sacrificed at the end of 4 w. [Results] Compared with the blank control group, the cas-pase-3 positive cells in each manganese group were significantly increased, the cyto-c positive cells were significantly decreased (P <0.01). Compared with 30mg / kg MnCl2 group and 15mg / kg MnCl2 group, the positive rate of caspase-3 was significantly increased and the rate of cyto-c positive cells was significantly decreased (P <0.01). The positive rates of caspase-3 and cyto-c positive cells in each group showed a significant negative correlation (r = -0.876, P <0.01). [Conclusion] Manganese can induce the apoptosis of rat spermatogenic cells through mitochondrial pathway, and the expression of caspase-3 increases with the increase of manganese dose. There is a certain dose-effect relationship between them.
