目的探讨氧化苦参碱对人胃癌SGC-7901细胞增殖的影响。方法分别应用2.0、1.5、1.0、0.5mg/mL氧化苦参碱体外培养人胃癌细胞SGC-7901,分别于培养24、48、72h后以倒置显微镜观察细胞形态学改变,采用MTT法观察不同浓度和作用时间下氧化苦参碱对SGC-7901细胞的抑制情况,并与对照组(不进行氧化若参碱处理)进行比较。结果 2.0 mg/mL浓度组氧化苦参碱作用人胃癌细胞株SGC-7901后24h抑制率[(35.500±0.014)%]高于1.5mg/mL组[(23.000±0.004)%]、1.0mg/mL组[(20.300±0.013)%]、0.5mg/mL[(15.700±0.007)%]和对照组(0),48h抑制率[(64.100±0.004)%]高于1.5mg/mL组[(45.400±0.013)%]、1.0mg/mL组[(44.700±0.031)%]、0.5mg/mL[(23.000±0.033)%]和对照组(0)(P<0.05),72h抑制率[(82.400±0.133)%]高于1.5 mg/mL组[(66.000±0.035)%]、1.0mg/mL组[(58.900±0.120)%]、0.5mg/mL[(34.900±0.024)%]和对照组(0)(P<0.05)。结论氧化苦参碱可抑制人胃癌细胞株SGC7901细胞增殖,细胞抑制率与氧化苦参碱浓度呈时间-剂量依赖性。
Objective To investigate the effects of oxymatrine on the proliferation of human gastric cancer cell line SGC-7901. Methods Human gastric cancer SGC-7901 cells were cultured with 2.0, 1.5, 1.0 and 0.5 mg / mL oxymatrine in vitro. The morphological changes of SGC-7901 cells were observed by inverted microscope 24, 48 and 72 hours after culture. Conclusion Oxymatrine can inhibit the proliferation of human gastric cancer cell line SGC7901 in a time-and dose-dependent manner with oxymatrine concentration.
