目的 :研究穿琥宁对内毒素 (LPS)诱导大鼠肺泡巨噬细胞 (AM )过度分泌肿瘤坏死因子 (TNF α)的影响及机制。方法 :体外培养大鼠AM ,分为对照组、LPS组、穿琥宁 +LPS组、来立信 +LPS组和穿琥宁 +来立信 +LPS组。测定上清TNF α水平、上清LPS水平以及细胞中I Κb的表达。并在电镜下观察穿琥宁对LPS结构的影响。结果 :经穿琥宁作用后 ,上清中的TNF α和LPS含量明显下降 ,而且电镜下证实LPS已失去其正常结构 ,同时 ,I Κb的表达也增加。结论 :穿琥宁对LPS诱导大鼠AM过度分泌TNF α有明显抑制作用。其机制可能与破坏内毒素结构和抑制I Κb的低表达有关
Objective: To study the effect and mechanism of Chuanhuning on excessive secretion of tumor necrosis factor (TNF α) from alveolar macrophages (AM) induced by endotoxin (LPS) in rats. METHODS: Rats were cultured in vitro and divided into control group, LPS group, Chuanhuning+LPS group, Tolixin+LPS group, and Chuanhuning+Likelixin+LPS group. Supernatant TNFα levels, supernatant LPS levels, and I Κb expression in cells were determined. The effect of Chuanhuning on LPS structure was observed under electron microscope. RESULTS: After the effect of Chuan Hu Ning, the content of TNF α and LPS in the supernatant decreased significantly, and under the electron microscope, LPS had lost its normal structure. At the same time, the expression of I Κ b also increased. Conclusion: Chuanhuning inhibited the excessive secretion of TNF α by AM induced by LPS in rats. The mechanism may be related to the destruction of endotoxin structure and inhibition of low expression of I Κb
