目的 :观察中等加热温度和低剂量放射对 MCF- 7单层细胞和细胞球的细胞杀伤作用。材料与方法 :通过测量 MCF- 7单层细胞及不同大小细胞球 (70μm、2 0 0μm、4 0 0μm、70 0μm)来观察细胞杀伤作用。用流式细胞仪来测量 4 1℃加热过程中的细胞周期变化 ,用克隆形成法来计算细胞生存率。结果 :MCF- 7细胞球 4 1℃加热后其生存率低于单层细胞 ,而 4 3℃加热后其生存率则高于单层细胞。 4 1℃加热过程中发生热耐受。同时本组实验发现 MCF- 7单层细胞和细胞球对放射敏感性相差不大。结论 :在4 1℃加热过程中 ,发生 G1阻滞 ,这与加热杀伤的敏感性有关
Objective : To observe the killing effect of medium heating temperature and low dose radiation on MCF-7 monolayer cells and cell spheres. Materials and Methods: Cell killing was observed by measuring MCF-7 monolayer cells and different size cell spheres (70 μm, 200 μm, 400 μm, 700 μm). Flow cytometry was used to measure cell cycle changes during heating at 1°C. Cell survival was calculated by cloning. RESULTS: The survival rate of MCF-7 cells after heated at 41 °C was lower than that of monolayer cells. The survival rate of MCF-7 cells after heating at 4 °C was higher than that of monolayer cells. 4 Thermal tolerance occurs during heating at 1°C. At the same time, this group of experiments found that MCF-7 monolayer cells and cell spheres have little difference in radiosensitivity. Conclusion: G1 blockade occurs during heating at 4 1°C, which is related to the sensitivity of heat killing
