目的:观察养心通脉颗粒Ⅱ方对气虚血瘀型心衰大鼠心肌细胞凋亡指数的影响。方法:通过多因素诱导复制中医气虚血瘀大鼠模型,通过阿霉素诱导复制心衰大鼠模型。60只SD雌性大鼠,随机分为养心通脉颗粒Ⅱ方组、依那普利组、模型组及空白对照组每组各15只。造模后养心通脉颗粒Ⅱ方组、依那普利组分别予养心通脉颗粒Ⅱ方、依那普利灌胃治疗,模型组、对照组予生理盐水灌胃治疗4周。末次给药后1h处死大鼠,取大鼠心脏,测大鼠左心重量指数,用tunel法检测心肌细胞凋亡指数。结果:与模型组对比养心通脉颗粒Ⅱ方组心肌细胞凋亡指数、左心重量指数明显减轻(P<0.05)。结论:养心通脉颗粒Ⅱ方可有效的抑制心肌细胞凋亡,从而减缓心衰的进展改善心衰症状,其作用机制与依那普利相似。
Objective: To observe the effects of Yangxin Tongmai Granule (Ⅱ) on the cardiomyocyte apoptosis index in rats with deficiency of qi and blood stasis. Methods: The model of Qi-Deficiency and Blood-Stasis of TCM was induced by multi-factor induction and the rat model of heart failure was induced by doxorubicin. Sixty female SD rats were randomly divided into Yangxintongmai Granule Ⅱ group, enalapril group, model group and blank control group, with 15 rats in each group. Yang Xin Tong Mai granule Ⅱ group and enalapril group were fed with Yangxintongmai Granule Ⅱ and Enalapril intragastrically after modeling respectively. The model group and the control group were treated with normal saline for 4 weeks. The rats were sacrificed at 1h after the last administration, the heart of rats was taken, the left heart weight index of rats was measured, and the apoptosis index of cardiomyocytes was detected by tunel method. Results: Compared with the model group, the myocardial apoptosis index and left ventricular mass index of Yangxintongmai Granule Ⅱ group were significantly reduced (P <0.05). CONCLUSION: Yangxintongmai Granule Ⅱ can effectively inhibit the apoptosis of cardiomyocytes, thereby slowing the progression of heart failure and improving the symptoms of heart failure. The mechanism of action is similar to enalapril.
