目的:考察2,3-二氢环戊二烯香豆素类化合物对何首乌毛状根生长的影响,并研究其对毛状根中有效成分二苯乙烯苷累积的刺激作用。方法:将2,3-二氢环戊二烯香豆素类化合物在不同的培养时间加入到毛状根中,共培养特定时间后测定其二苯乙烯苷的含量及生长量,并通过考察不同浓度和继代次数确定最佳实验条件。结果:加入香豆素类化合物的最佳时间为预培养第4天,最佳质量浓度为0.025 g.L-1,毛状根的生长量有较大提高,与对照组相比二苯乙烯苷的含量增加4倍左右。结论:2,3-二氢环戊二烯香豆素类化合物作为一类非生物诱导子能够促进何首乌毛状根的生长及有效成分二苯乙烯苷的累积。
OBJECTIVE: To investigate the effects of 2,3-dihydrocyclopentadienyl coumarins on the growth of hairy root of Polygonum multiflorum and to study its stimulating effect on the accumulation of stilbene glycoside in hairy root. Methods: The 2,3-dihydrocyclopentadienyl coumarins were added to the hairy roots at different culture time. After co-cultured for a certain period of time, their content and growth of stilbene glycoside were determined. Different concentrations and subcultures determine the best experimental conditions. Results: The optimum time for the addition of coumarins was on the 4th day of pre-culture, the best mass concentration was 0.025 gL-1, and the growth of hairy roots was greatly increased. Compared with the control group, the content of stilbene glycosides The content increased about 4 times. CONCLUSION: 2,3-Dihydrocyclopentadienyl coumarins can promote the growth of hairy root of Polygonum multiflorum and the accumulation of stilbene glucoside as a kind of non-biological elicitor.
