NF-κB通过结合microRNA-130a启动子抑制其靶基因PTEN调控宫颈癌细胞增值

  NF-κB能够激活一系列控制细胞信号通路的基因,其产物是多个生物过程的重要组成部分.microRNA是一系列内源非编码RNA,能够调节基因表达,并在肿瘤发生过程中扮演重要角色.通过对人宫颈癌细胞系进行研究,我们探索了NF-κB是否能够通过结合miR-130a的启动子区域来调控其表达;我们发现miR-130a可通过靶向PTEN促进宫颈癌细胞生长;此外,NF-κB可通过上调miR-130a的表达来促进癌细胞系CaSki和HeLa生长;miR-130a可通过直接靶向PTEN基因的3端非翻译区促进细胞生长,抑制AKT通路活性.综上,我们的研究为宫颈癌中NF-κB/miR-130a/PTEN通路的作用提供了新的视角,也为宫颈癌治疗提供了新的潜在的治疗靶点.
