目的观察转化生长因子β_1(TGF-β_1)、叉头框蛋白Q1(Fox Q1)、E-钙黏蛋白(E-cad)和N-钙黏蛋白(N-cad)、血管内皮生长因子(VEGF)在上皮性卵巢癌卵巢上皮组织中的表达,并探讨其意义。方法上皮性卵巢癌40例(恶性组)、良性上皮性卵巢肿瘤39例(良性组)、正常卵巢38例(正常组),手术切取卵巢上皮组织,采用实时荧光定量PCR和免疫组织化学法分别检测各组卵巢上皮组织中TGF-β_1、FOXQ1、E-cad、N-cad、VEGF的mRNA和蛋白表达。采用Spearman相关分析法分析上述各指标之间的相关性。结果恶性组TGF-β_1、FOXQ1、N-cad、VEGF mRNA、蛋白表达高于正常组和良性组,E-cadmRNA、蛋白表达低于正常组、良性组(P均<0.05)。上皮性卵巢癌中TGF-β_1、FOXQ1、E-cad、N-cad、VEGF表达与FIGO分期、淋巴结转移有关。上皮性卵巢癌中TGF-β_1与FOXQ1、E-cad、N-cad呈正相关(r分别为0.335、0.123、0.411,P分别为0.003、0.002、0.000);FOXQ1与E-cad呈正相关(r=0.321,P=0.000);E-cad与N-cad、VEGF呈正相关(r分别为0.433、0.283,P分别为0.000、0.010)。结论上皮性卵巢癌卵巢上皮组织中TGF-β_1、FOXQ1、VEGF、N-cad表达升高,E-cad表达降低,共同参与上皮性卵巢癌的发生发展、侵袭及转移。
Objective To investigate the expression of TGF-β_1, Fox Q1, E-cad and N-cadherin, vascular endothelial growth factor ) In epithelial ovarian epithelial ovarian epithelial tissue and to explore its significance. Results The mRNA and protein expressions of TGF-β 1, FOXQ1, N-cad and VEGF in malignant group were significantly higher than those in normal group and benign group (P <0.05). The expression of TGF-β1, FOXQ1, E-cad, N-cad and VEGF in epithelial ovarian cancer is related to FIGO stage and lymph node metastasis. The positive rates of FOXQ1, E-cad and N-cad in epithelial ovarian cancer were positively correlated (r = 0.335,0.123,0.411, P = 0.003,0.002,0.000, respectively) 0.321, P = 0.000). There was a positive correlation between E-cad and N-cad and VEGF (r = 0.433,0.283, P = 0.000,0.010, respectively). Conclusions The expression of TGF-β 1, FOXQ1, VEGF and N-cad in epithelial ovarian epithelial tissue of epithelial ovarian cancer is increased, and the expression of E-cad is decreased. It is involved in the occurrence, development, invasion and metastasis of epithelial ovarian cancer.
