目的:本课题是为了深入研究采用助阳通便汤治疗便秘的作用机理,进而揭示助阳通便汤治疗便秘的可能作用机制,为中医治疗慢传输型便秘提供实验依据。方法:将112只小鼠随机分成正常组、模型组、助阳通便汤低剂量组、助阳通便汤中剂量组、助阳通便汤高剂量组、麻仁软胶囊组、西沙比利组,共7组,每组16只,雌雄各半。除正常组外,其余各组均采用皮下注射盐酸吗啡的方法,复制出慢传输型便秘小鼠模型,给予相应的药物灌胃,观察助阳通便汤对便秘模型小鼠的通便作用,应用免疫组化的方法研究本复方对便秘模型小鼠结肠肌间神经丛血管活性肠肽(VIP)和5-羟色胺(5-HT)受体含量的影响。结果:助阳通便汤治疗组小鼠结肠组织中5-HT受体和VIP阳性表达细胞分布相对较密集。平均光密度值增高。结论:助阳通便汤能增加便秘小鼠结肠5-HT受体和VIP阳性细胞数量。助阳通便汤治疗慢传输型便秘的作用机制之一是通过调节结肠组织中神经递质(5-HT受体和VIP)的含量来实现的。
Objective: The purpose of this project is to further study the mechanism of treating nausea and relieving constipation by using nourishing yang and purging constipation, and to reveal the possible mechanism of nourishing yang and purging constipation to treat constipation. Methods: One hundred and twelve mice were randomly divided into four groups: normal group, model group, low-dose group of nourishing and catharsis group, middle-dose group of nourishing and catharsis group, high-dose group of nourishing and catharsis group, soft capsule group of Miren group, Lee group, a total of 7 groups, 16 in each group, male and female. In addition to the normal group, the rest of the groups were subcutaneous injection of morphine hydrochloride method to copy a slow-transit constipation mouse model, given the corresponding drug gavage to observe the constipation model to help purgative effect of catharsis, Immunohistochemistry was used to study the effect of this compound on the contents of VIP and 5-HT in colon myenteric plexus of constipation model mice. RESULTS: The distribution of 5-HT receptor and VIP positive cells in the colon of mice in the recipe of yang yan tong shou tang was relatively dense. Average optical density increased. Conclusion: Zhuiyong Tongbang Decoction can increase the number of colonic 5-HT receptor and VIP positive cells in constipation mice. One of the mechanisms by which Jiyang Tongbang Decoction is used to treat slow transit constipation is through regulating the contents of neurotransmitters (5-HT receptor and VIP) in colon tissue.
