以越橘果实转录组测序文库为基础,借助生物信息学方法对越橘PDR型转运蛋白基因进行鉴定,并利用实时荧光定量PCR技术对其在越橘不同器官和组织中的相对表达量进行分析。结果表明:越橘果实至少含有30个PDR转运蛋白基因;从越橘果皮和果肉差异表达文库中筛选出21个差异表达基因,且都是在果皮中上调表达的基因;通过qRT-PCR分析,发现21个PDR转运蛋白基因在越橘不同器官和组织中的表达具有组织特异性,其中有16个基因在根中表达量最高。
Based on the sequence library of cranberry transcriptome, the gene of bilberry PDR-type transporter was identified by bioinformatics method and its relative expression in different organs and tissues of bilberry was analyzed by real-time fluorescence quantitative PCR . The results showed that there were at least 30 PDR transporter genes in bilberry fruit, 21 differentially expressed genes were screened from differential expression libraries of bilberry pericarp and flesh, and all of them were up-regulated genes in pericarp. By qRT-PCR analysis, Twenty-one PDR transporter genes were found to be tissue-specific in different organs and tissues of bilberry, of which 16 were highest in roots.
