目的:研究制备抗人喉癌/抗蓖麻毒素A双功能抗体用以导向喉癌的治疗。方法:在制备抗人喉癌单克隆抗体杂交瘤细胞基础上,通过二次杂交瘤技术制备能够分泌抗人喉癌和抗蓖麻毒素A的杂交瘤细胞株。结果:该双功能抗体对Hep-2细胞株杀伤力比化学偶联法的双功能抗体强15倍。结论:采用二次杂交瘤技术制备的双功能抗体比化学偶联法的细胞毒性作用显著增高,优于化学偶联法。
Objective: To study the preparation of anti-human laryngeal cancer/anti-ricin A diabodies for the treatment of laryngeal cancer. METHODS: Hybridoma cell lines capable of secreting anti-human laryngeal carcinoma and anti-ricin A were prepared by hybridoma technique based on the preparation of anti-human laryngeal cancer monoclonal antibody hybridoma cells. RESULTS: The killing power of the bifunctional antibody against Hep-2 cell strain was 15 times stronger than that of the chemically coupled bifunctional antibody. Conclusion: The bifunctional antibody produced by the second hybridoma technique has significantly higher cytotoxicity than the chemical coupling method and is superior to the chemical coupling method.
