蛹虫草Cordyceps militaris是当前用于功能食品和药物开发上较受国内外关注的真菌之一。本研究通过1次一因素法和正交实验设计优化了蛹虫草产虫草菌素和菌丝体的深层培养工艺。最适合菌丝生长的pH值和温度为6.0和20℃,而最适合虫草素积累的pH值和温度为5.0和26℃。蔗糖、蛋白胨、MgSO4、K2HPO4和NAA分别是最适合虫草素积累的碳源、氮源、无机盐和生长因子;培养基的成分对蛹虫草生物量的影响次序为:蔗糖>蛋白胨>MgSO4·7H2O>K2HPO4>NAA;培养基的成分对蛹虫草产虫草素的影响次序为:蛋白胨>K2HPO4>NAA>MgSO4·7H2O>蔗糖。最适合蛹虫草菌丝生长的培养基配比为4%蔗糖,2.5%蛋白胨,0.05%MgSO4·7H2O,0.15%K2HPO4,2.0mg/l NAA;最适合蛹虫草深层发酵产虫草素的培养基配比为4%蔗糖,1.5%蛋白胨,0.05%MgSO4·7H2O,0.05%K2HPO4,4.0mg/l NAA的培养基;在最佳蛹虫草深层发酵产虫草素的工艺条件下,CM8菌株胞外虫草菌素产量达到961.21 mg/l,胞内虫草菌素的含量达到12.87mg/g;菌丝体的产量为22.97 g/l,本研究得出的结论对深层培养提高蛹虫草虫草菌素和菌丝体的产量有借鉴意义。

The most suitable culture medium for the growth of Cordyceps militaris mycelia was 4% sucrose, 2.5% peptone, 0.05% MgSO4 · 7H2O, 0.15% K2HPO4 and 2.0 mg / l NAA. Culture medium containing 4% sucrose, 1.5% peptone, 0.05% MgSO 4 · 7H 2 O, 0.05% K 2 HPO 4 and 4.0 mg / l NAA. Under the optimal fermentation conditions of Cordycepin, The yield of mycotoxin reached 12.87mg / g and the yield of mycelia was 22.97g / l.
