目的:探讨植物激素中吲哚乙酸/辣根过氧化物酶(IAA/HRP)对人涎腺腺样囊性癌SACC-83细胞体外增殖和凋亡的影响及机制。方法:采用CCK-8比色法检测IAA/HRP对人涎腺腺样囊性癌SACC-83增殖的抑制作用,应用实时荧光定量PCR及Western blot检测凋亡相关基因Caspase-3和Livinα基因mRNA及蛋白表达水平的改变。结果:IAA/HRP对SACC-83细胞增殖有明显的抑制作用,且呈时间和浓度依赖性(P<0.05);不同浓度的IAA/HRP(60、100μmol/L IAA+1.2 mg/L HRP)可上调Caspase-3 mRNA及蛋白的表达(P<0.05),下调LivinαmRNA及蛋白的表达(P<0.05)。结论:IAA/HRP可抑制SACC-83增殖、诱导细胞凋亡,其作用机制是通过调节Caspase-3和Livinα的表达而实现的。
AIM: To investigate the effects and mechanisms of IAA / HRP on proliferation and apoptosis of human salivary adenoid cystic carcinoma SACC-83 cells in vitro. Methods: The inhibitory effect of IAA / HRP on the proliferation of human salivary adenoid cystic carcinoma SACC-83 cells was detected by CCK-8 colorimetric assay. The mRNAs of apoptosis-related genes Caspase-3 and Livinα were detected by real-time fluorescence quantitative PCR and Western blot And protein expression level changes. Results: IAA / HRP significantly inhibited the proliferation of SACC-83 cells in a time and concentration dependent manner (P <0.05). IAA / HRP (60,100 μmol / L IAA and 1.2 mg / L HRP) The mRNA and protein expression of Caspase-3 were up-regulated (P <0.05), and the expression of Livinα mRNA and protein was down-regulated (P <0.05). Conclusion: IAA / HRP can inhibit the proliferation and induce apoptosis of SACC-83 cells through regulating the expression of Caspase-3 and Livinα.
