目的观察葛根素联合曲美他嗪对大鼠心肌缺血再灌注损伤的作用机制。方法将48只SD大鼠随机分为对照组、模型组、曲美他嗪组、葛根素联合曲美他嗪组(联合组),每组12只。对照组不结扎左冠状动脉前降支,模型组、曲美他嗪组、联合组结扎左冠状动脉前降支90 min,松解120 min制备急性心肌梗死再灌注模型,并于再灌注结束即刻,依次由尾静脉分别注射生理盐水、曲美他嗪、葛根素联合曲美他嗪,再灌注120 min后,取血检测心肌过氧化物酶、丙二醛和血清肌酸激酶水平,并观察心电图ST段抬高指数。结果联合组、曲美他嗪组血清肌酸激酶、过氧化物酶和丙二醛活性低于模型组(P<0.01),ST段抬高指数低于模型组(P<0.05);联合组血清肌酸激酶活性低于曲美他嗪组(P<0.05)。结论葛根素联合曲美他嗪可减轻SD大鼠心肌缺血再灌注损伤。
Objective To observe the mechanism of puerarin combined with trimetazidine on myocardial ischemia-reperfusion injury in rats. Methods 48 SD rats were randomly divided into control group, model group, trimetazidine group, puerarin and trimetazidine group (combination group), 12 in each group. In the control group, the left anterior descending coronary artery, the model group and the trimetazidine group were not ligated. The anterior descending branch of the left coronary artery was ligated for 90 min in the combined group and the model of acute myocardial infarction reperfusion was prepared by releasing for 120 min. At the end of reperfusion , Followed by the tail vein injection of saline, trimetazidine, puerarin and trimetazidine, 120 min after reperfusion, blood was collected to detect myocardial peroxidase, malondialdehyde and serum creatine kinase levels and observed ECG ST segment elevation index. Results Serum creatine kinase, peroxidase and malondialdehyde in the combination group and the trimetazidine group were lower than those in the untreated group (P <0.01), and the ST segment elevation index was lower than that in the model group (P <0.05) Serum creatine kinase activity was lower in the trimetazidine group (P <0.05). Conclusion Puerarin combined with trimetazidine can attenuate myocardial ischemia-reperfusion injury in SD rats.
