目的探讨重庆地区汉族人群肽酰精氨酸脱亚胺基酶4(peptidylarginine deiminase4,PADI4)单核苷酸多态性(SNP)与类风湿关节炎(rheumatoid arthritis,RA)之间的关系。方法采用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)法检测302例RA患者和322名健康对照者的PADI4rs874881SNP多态性,同时检测类风湿因子(Rheumatoidfactor,RF)、抗环瓜氨酸肽抗体(cyclic citrullinated peptide,CCP)和抗角蛋白抗体(anti-keratin antibody,AKA)3种RA的特异性自身抗体。结果健康对照组PADI4rs874881C/C、C/G和G/G基因型频率分别为0.332、0.488和0.180;RA组分别为0.295、0.474和0.232,与对照组比较,差异无统计学意义(P>0.05)。RA组RF、抗CCP抗体和抗AKA抗体检测阳性率、滴度水平与PADI4rs874881基因分型均无相关性(P>0.05)。结论重庆汉族人群PADI4rs874881多态性与RA的易感性无关,与RF、CCP及AKA等抗体的阳性率无相关性。
Objective To investigate the relationship between peptidylarginine deiminase 4 (PADI4) single nucleotide polymorphism (SNP) and rheumatoid arthritis (RA) in Han population of Chongqing. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the polymorphisms of PADI4 rs874881 SNP in 302 RA patients and 322 healthy controls. Rheumatoid factor (RF) , Cyclic citrullinated peptide (CCP) and anti-keratin antibody (AKA). Results The frequencies of PADI4 rs874881C / C, C / G and G / G genotypes were 0.332, 0.488 and 0.180 respectively in RA group and 0.295, 0.474 and 0.232 respectively in RA group, but there was no significant difference between the two groups (P> 0.05 ). The positive rates of RF, anti-CCP and anti-AKA antibodies in RA group were not correlated with the PADI4 rs874881 genotype (P> 0.05). Conclusion The polymorphism of PADI4 rs874881 in Chongqing Han population has nothing to do with the susceptibility of RA, and no correlation with the positive rate of antibodies such as RF, CCP and AKA.
