用外科切除的人肝癌组织抽提的膜抗原和细胞悬液免疫动物,细胞融合后筛选出一侏抗人原发性肝细胞癌单克隆抗体,命名Hepatoma Antibody.5,(HAb_5)。此抗体对50例临床病理诊断为肝癌的组织切片作免疫组织化学定位诊断,阳性率为88%,7例肝转移性腺癌染色,均无交叉反应,故认为对诊断和鉴别原发性肝癌与转移性肝癌方面有一定实用价值。为进一步确定HAb_5(?)相应抗原的性质,用Sephadex-G200纯化HAb_5腹水抗体。用Sepharose 4B制备免疫亲合层析柱纯化HAb_5相应抗原。经SDS-PAGE和免疫斑点(ABC)法,高碘酸氧化试验初步结果表明,HAb_5抗原是一种分子量为68000的糖蛋白。此抗原物质的血清学诊断意义有待进一步研究。
The animals were immunized with membrane antigen and cell suspension extracted from surgically excised human hepatocellular carcinoma tissue. After confluence, a monoclonal antibody against human primary hepatocellular carcinoma was screened and named Hepatoma Antibody.5 (HAb_5). This antibody was used in 50 cases of histologically diagnosed hepatocellular carcinoma for immunohistochemical localization. The positive rate was 88%. There were no cross reactions in 7 cases of hepatic metastatic adenocarcinoma. Therefore, it is of great value for the diagnosis and differential diagnosis of primary hepatocellular carcinoma Metastatic liver cancer has some practical value. To further determine the nature of the HAb_5 (?) Antigen, HAb_5 ascites was purified with Sephadex-G200. Sepharose 4B was used to prepare the corresponding antigen of HAb_5 by immunoaffinity chromatography. Preliminary results of the periodate oxidation test by SDS-PAGE and immunofluorescence (ABC) showed that the HAb_5 antigen was a glycoprotein with a molecular weight of 68,000. Serological diagnostic significance of this antigenic substances needs further study.
