目的:分析富含亮氨酸的三角状五肽重复结构蛋白(LRPPRC)和抗凋亡蛋白Bcl-2在前列腺癌组织的表达及LRPPRC对前列腺癌细胞凋亡的影响。方法:应用免疫组化法检测198例前列腺癌组织中LRPPRC和Bcl-2的表达;分析LRPPRC和Bcl-2表达的相关性;分析LRPPRC和Bcl-2表达与预后的关系;应用si RNA LRPPRC转染前列腺癌细胞下调LRPPRC水平,观察对细胞增殖、凋亡和Bcl-2、Bax、caspase-3蛋白表达的影响。结果:前列腺癌和前列腺增生组织LRPPRC的表达率分别为61.6%(122/198)和18.0%(18/100)(P<0.001);Bcl-2的表达率分别为49.5%(98/198)和45.0%(45/100)(P=0.539)。LRPPRC和Bcl-2的表达有显著正相关性(r=0.179,P=0.012)。LRPPRC和Bcl-2同时高表达的患者无疾病进展生存和总生存期均显著降低(P<0.001)。下调LRPPRC表达可抑制前列腺癌细胞增殖,促进前列腺癌细胞凋亡,降低Bcl-2蛋白的表达,使caspase-3活化增多,对Bax表达无显著影响。结论:前列腺癌组织LRPPRC表达显著增高;LRPPRC与抑制凋亡有关;LRPPRC在前列腺癌中的作用机制与抗凋亡蛋白Bcl-2有关。
OBJECTIVE: To analyze the expression of leptin-rich trigonal pentagonal repeat structural protein (LRPPRC) and anti-apoptotic protein Bcl-2 in prostate cancer and the effect of LRPPRC on the apoptosis of prostate cancer cells. METHODS: The expressions of LRPPRC and Bcl-2 in 198 cases of prostate cancer were detected by immunohistochemistry. The correlation between LRPPRC and Bcl-2 expression was analyzed. The relationship between LRPPRC and Bcl-2 expression and prognosis was analyzed. Proliferation of prostate cancer cells down-regulated LRPPRC levels were observed on cell proliferation, apoptosis and Bcl-2, Bax, caspase-3 protein expression. Results: The expression rates of LRPPRC in prostate cancer and benign prostatic hyperplasia tissues were 61.6% (122/198) and 18.0% (18/100) respectively (P <0.001), while the expression rates of Bcl-2 were 49.5% (98/198) And 45.0% (45/100) (P = 0.539). There was a significant positive correlation between LRPPRC and Bcl-2 expression (r = 0.179, P = 0.012). Patients with both LRPPRC and Bcl-2 overexpression had significantly lower progression-free survival and overall survival (P <0.001). Down-regulation of LRPPRC could inhibit the proliferation of prostate cancer cells, promote the apoptosis of prostate cancer cells, decrease the expression of Bcl-2 protein, increase the activation of caspase-3, but had no significant effect on the expression of Bax. Conclusion: The expression of LRPPRC in prostate cancer tissues is significantly increased. LRPPRC is associated with the inhibition of apoptosis. The mechanism of LRPPRC in prostate cancer is related to the anti-apoptotic protein Bcl-2.
