目的 探讨大鼠脑缺血再灌注后血脑屏障 (BBB)通透性的改变以及转移生长因子 β1(TGF β1 )在脑组织中的表达。方法 采用线栓法制备大鼠局灶性脑缺血再灌注模型 ,通过测定脑组织中伊文氏蓝 (EB)含量及免疫组化法来观察TGF β1 的表达。结果 缺血 2h再灌注 3h ,BBB通透性开始增加 ,2 4h达高峰 ,72h后明显减弱。同时 ,TGF β1 在缺血再灌注 3h开始表达 ,2 4h达高峰 ,持续至 72h ,72h后逐渐减弱。结论 脑缺血后BBB的破坏与TGF β1 的表达密切相关 ,提示TGF β1 参与了BBB内皮细胞破坏的修复过程
Objective To investigate the changes of permeability of the blood-brain barrier (BBB) and the expression of transforming growth factor-β1 (TGF-β1) in brain after cerebral ischemia-reperfusion in rats. Methods Focal cerebral ischemia-reperfusion model was established by thread occlusion method. The expression of TGF-β1 was observed by measuring the content of Evans blue (EB) in brain tissue and immunohistochemistry. RESULTS: After 2h of ischemia and 3h of reperfusion, the permeability of BBB began to increase, reached the peak at 24 hours and decreased significantly after 72 hours. At the same time, TGFβ1 began to be expressed at 3h after ischemia-reperfusion and peaked at 24h, and continued to 72h and gradually decreased after 72h. Conclusions The damage of BBB after cerebral ischemia is closely related to the expression of TGF-β1, suggesting that TGF β1 is involved in the repair process of BBB endothelial cell destruction
