目的 在体条件下观察 β 淀粉样蛋白 (Aβ)对白细胞介素 1β(IL 1β)和神经组织蛋白(S10 0 β)表达的影响 ,探讨三者之间的相互关系。方法 以不同浓度Aβ溶液及生理盐水作海马立体定向注射后 ,采用免疫组织化学、HE染色及刚果红染色方法 ,显示胶质细胞反应及IL 1β、S10 0 β表达情况 ,并在高倍显微镜下 (× 40 0 )进行阳性细胞计数 ,方差分析和等级相关分析作显著性检验。结果 胶质细胞反应程度及IL 1β、S10 0 β的表达水平随Aβ浓度增大而增加 ,4μg μlAβ溶液注射组IL 1β、S10 0 β阳性细胞数 (个 视野 )分别为 41.75± 2 .6 1和 42 .39± 3.46 ,显著多于生理盐水组 (2 1.16±2 .92和 34 .5 0± 3.5 7) ,P <0 .0 1;S10 0 β+ 与IL 1β+ 细胞数存在较强的相关性 (rs=0 .892 )。结论 在体条件下高含量Aβ强烈诱导胶质细胞IL 1β和S10 0 β的表达 ,结果提示三者之间的相互作用在Alzheimer病发病机制中有重要作用。
Objective To observe the effect of amyloid β (Aβ) on the expression of interleukin 1β (IL 1β) and neural tissue protein (S10 0 β) in vivo and to explore the relationship among the three. Methods The hippocampal injection of Aβ solution and saline at different concentrations was used for stereotaxic injection. Immunohistochemistry, HE staining and Congo red staining were used to observe glial cell responses and IL 1β and S10 0 β expression. × 40 0) for positive cell counts, analysis of variance and rank correlation analysis for significant test. Results The glial cell response and the expression levels of IL 1β and S10 0 β increased with the increase of Aβ concentration. The number of IL 1β and S10 0 β-positive cells in 4 μg μl Aβ injection group was 41.75 ± 2. And 42.39 ± 3.46, respectively, which were significantly higher than those in the saline group (2 1.16 ± 2.92 and 34.500 ± 3.57), P <0.01; the number of S10 0β + and IL 1β + cells was significantly higher (Rs = 0.882). CONCLUSION: High levels of Aβ under in vivo conditions strongly induce the expression of IL-1β and S10 0β in glial cells. The results suggest that the interaction between the three plays an important role in the pathogenesis of Alzheimer’s disease.
